ABSTRACT

Abstract Neospora caninum is an apicomplexan parasite that causes abortion in cattle, resulting in significant economic losses. There is no commercial treatment for neosporosis, and drug repositioning is a fast strategy to test possible candidates against N. caninum. In this article, we describe the effects of atovaquone, chloroquine, quinine, primaquine and tetracycline on N. caninum proliferation. The IC50 concentrations in N. caninum were compared to the current information based on previous studies for Plasmodium and Toxoplasma gondii, correlating to the described mechanisms of action of each tested drug. The inhibitory patterns indicate similarities and differences among N. caninum, Plasmodium and T. gondii. For example, atovaquone demonstrates high antiparasitic activity in all the analyzed models, while chloroquine does not inhibit N. caninum. On the other hand, tetracycline is effective against Plasmodium and N. caninum, despite its low activity in T. gondii models. The repurposing of antimalarial drugs in N. caninum is a fast and inexpensive way to develop novel formulations using well-established compounds.


Resumo Neospora caninum é um parasita Apicomplexa relacionado a abortos no gado bovino, que resultam em impactos econômicos. Não há tratamento comercial para neosporosis e o reposicionamento de drogas indica uma estratégia rápida para testar candidatos anti-N. caninum. Neste artigo, são descritos os efeitos da atovaquona, cloroquina, quinino, primaquine e tetraciclina na proliferação de N. caninum. As concentrações IC50 em N. caninum foram comparadas com a informação disponível, baseada em estudos publicados previamente para Plasmodium e Toxoplasma gondii, incluindo a correlação com os mecanismos de ação descritos para cada droga testada. Os padrões de inibição indicam pontos de similaridades e diferenças entre N. caninum, Plasmodium e T. gondii. Por exemplo, a atovaquona demonstra uma alta atividade antiparasitária em todos os modelos testados, enquanto a cloroquina não inibe N. caninum. Por outro lado, a tetraciclina é efetiva contra Plasmodium e N. caninum, em contraste com a baixa atividade em modelos de T. gondii. O reposicionamento de drogas antimaláricas em N. caninum é uma forma rápida e de baixo custo para o desenvolvimento de novas formulações que usam compostos bem estabelecidos.

OBJECTIVES@#To analyze the chromatic properties and translucency of porcelain veneers made from different ceramic materials against the background of tetracycline-stained teeth.@*METHODS@#Porcelain specimens (A1, A3, B2, B4) measuring 0.50 mm in thickness were prepared by heat-press casting and layering. The L*, a*, and b* values of the specimens against simulated tetracycline tooth and black-and-white backgrounds were measured by a spectrophotometer, and color differences ΔE@*RESULTS@#The ΔE@*CONCLUSIONS@#When changing the color of tetracycline-stained teeth, 0.50 mm-thick IPS d.SIGN feldspathic veneers with an opaque layer provide better chromatic properties than IPS e.max Press LT glass ceramic veneers. However, the translucency of feldspathic veneers is generally poorer than that of glass ceramic veneers.

OBJECTIVES@#This study aimed to evaluate the color stability of tetracycline teeth restored with ceramic veneers of different thicknesses combined with different resin cement systems after aging.@*METHODS@#Twenty patients with tetracycline teeth, including two maxillary central incisors, were selected clinically. The patients were randomly divided into four groups and restored with 0.5 and 0.75 mm ceramic veneers by using a veneer adhesive system, either with light-cured or dual-cured reaction. The color difference (ΔE) values after cementation and 1, 6, 12, and 24 months of use were obtained by quantification of L*, a*, and b* values with a colorimeter. The results were analyzed statistically with two-way ANOVA and Student's @*RESULTS@#The ΔE values of ceramic veneers detected after aging were less than 2.25. The 0.5 mm groups exhibited greater color change than the 0.75 mm-thick veneers (@*CONCLUSIONS@#Resin cements and veneer thickness influence the color of ceramic veneers after aging. Cementation of veneers with either dual- or light-cured resin cements does not affect the long-term color stability of tetracycline teeth differently.

ABSTRACT

O Informe Diário de Evidências é uma produção do Ministério da Saúde que tem como objetivo acompanhar diariamente as publicações científicas sobre tratamento farmacológico e vacinas para a COVID-19. Dessa forma, são realizadas buscas estruturadas em bases de dados biomédicas, referentes ao dia anterior desse informe. Não são incluídos estudos pré-clínicos (in vitro, in vivo, in silico). A frequência dos estudos é demonstrada de acordo com a sua classificação metodológica (revisões sistemáticas, ensaios clínicos randomizados, coortes, entre outros). Para cada estudo é apresentado um resumo com avaliação da qualidade metodológica. Essa avaliação tem por finalidade identificar o grau de certeza/confiança ou o risco de viés de cada estudo. Para tal, são utilizadas ferramentas já validadas e consagradas na literatura científica, na área de saúde baseada em evidências. Cabe ressaltar que o documento tem caráter informativo e não representa uma recomendação oficial do Ministério da Saúde sobre a temática. Foram encontrados 16 artigos e 13 protocolos.

The evolution of bacterial resistance is generating a serious public health problem due to the indiscriminate use of antibiotics, the application of non-optimal doses, the irregularity in the taking of medicines sent by the health professional, factors that have affected the increase in the rate of antimicrobial resistance; It is important to generate strategies that contribute to diminishing it, including the rational use of antibiotics and the constant research of new therapeutic alternatives such as teixobactin, which is a product of the Gram negative bacterium called Eleftheria terrae, related to the genus Aquabacterium, is a microorganism that presents extremophile conditions, for which, a multichannel system of semipermeable membranes called Ichip was developed for its isolation. Eravacycline is a new fully synthetic bacteriostatic antibiotic of the tetracycline family, is a potent inhibitor based on the mechanism of the bacterial ribosome and exerts potent activity against a broad spectrum of susceptible and multiresistant bacteria.


La evolución de la resistencia bacteriana ha generado un serio problema de salud pública debido al uso indiscriminado antibioticos, la aplicación de dosis no óptimas, la irregularidad en la toma de medicinas prescritas por el profesional de la salud han llevado a un aumento en la tasa de resistencia antimicrobiana; por ello es importante generar estrategias que contribuyan a disminuirla incluyendo el uso racional de anitibioticos y la búsqueda constante de nuevos antibioticos. La teixobactina es un producto de bacterias gram negativas llamadas Eleftheria terrae, relacionadas con el género Aquabacterium, el cual es un microorganismo que presenta condiciones extremofilas. Para el aislamiento de este nuevo compuesto se utilizó un sistema multicanal de membranas semipepermeables llamadas Ichip. Eravaciclina es un nuevo antibiotico síntetico bacteriostatico de la familia de las tetraciclinas y es un potente inhibidor de la maquinaria ribosomal bacteriana, con una potente actividad contra un amplio espectro de bacterias multirresistentes.

La instrumentación mecánica durante el tratamiento periodontal trae consigo la formación de escombros microcristalinos que inhiben la adhesión tisular a la superfi cie radicular y favorece la proliferación bacteriana, lo cual perjudica los resultados del tratamiento periodontal a corto y largo plazo. Hoy en día el acondicionamiento radicular con el uso de biomodifi cadores es una opción de tratamiento adicional en el tratamiento de la periodontitis y el tratamiento de cobertura radicular. El objetivo del presente estudio es realizar una revisión de la literatura acerca de las aplicaciones y del acondicionamiento radicular con ácido cítrico, tetraciclina, EDTA y láser en el tratamiento periodontal no quirúrgico y quirúrgico (AU)


Mechanical instrumentation during periodontal treatment brings the formation of microcrystalline debris that inhibits tissue adhesion to the root surface and favors bacterial proliferation, which harms the results of the short and long term periodontal treatment. Nowadays, root conditioning with the use of biomodifi cators is an additional treatment option in the treatment of periodontitis and root coverage therapy. ABSTRACT

Although Mycoplasma pneumoniae pneumonia (MPP) has been generally susceptible to macrolides, the emergence of macrolide-resistant MPP (MRMP) has made its treatment challenging. MRMP rapidly spread after the 2000s, especially in East Asia. MRMP is more common in children and adolescents than in adults, which is likely related to the frequent use of macrolides for treating M. pneumoniae infections in children. MRMP is unlikely to be related to clinical, laboratory, or radiological severity, although it likely prolongs the persistence of symptoms and the length of hospital stay. Thereby, it causes an increased burden of the disease and poor quality of life for the patient as well as a societal socioeconomic burden. To date, the only alternative treatments for MRMP are secondary antimicrobials such as tetracyclines (TCs) or fluoroquinolones (FQs) or systemic corticosteroids; however, the former are contraindicated in children because of concerns about potential adverse events (i.e., tooth discoloration or tendinopathy). A few guidelines recommended TCs or FQs as the second-line drug of choice for treating MRMP. However, there have been no evidence-based guidelines. Furthermore, safety issues have not yet been resolved. Therefore, this article aimed to review the benefits and risks of therapeutic alternatives for treating MRMP in children and review the recommendations of international or regional guidelines and specific considerations for their practical application.

ABSTRACT

El aumento de la resistencia bacteriana múltiple a antimicrobianos es considerado una gran amenaza para la salud pública mundial y como generador de una importante crisis en el funcionamiento de los sistemas de salud. Esta crisis es discutida diariamente por los gobiernos y los parlamentos, las instituciones globales de salud, fundaciones benéficas y de científicos y de profesionales de la salud y también de consumidores de productos animales. En todos los países del orbe se ha identificado al uso de antimicrobianos en la crianza industrial de animales como un importante determinante en la selección de esta resistencia. Aprovechando la oportunidad que se ha planteado en Chile con el diseño del Plan Nacional Contra la Resistencia a los Antimicrobianos, hemos creído importante revisitar y actualizar sumariamente nuestros estudios sobre el uso de antimicrobianos en la acuicultura del salmón y de su potencial impacto en el ambiente y la salud humana y animal. Estos estudios indican que 95% de tres grupos de antimicrobianos importados al país, que incluyen tetraciclinas, fenicoles y quinolonas, son usados en medicina veterinaria y mayormente en la acuicultura del salmón. Nuestros estudios indican que el excesivo uso de estos antimicrobianos genera la presencia de residuos de antimicrobianos en el ambiente marino hasta 8 km de los sitios de acuicultura, los que seleccionan a bacterias con resistencia múltiple en dicho ambiente, ya que ellas contienen variados genes de resistencia a estos antimicrobianos. Estos genes de resistencia están contenidos en elementos genéticos móviles incluyendo plásmidos e integrones, los que son trasmitidos a otras bacterias permitiendo su potencial diseminación epidémica entre poblaciones bacterianas. Bacterias del ambiente marino contienen genes idénticos a los genes de resistencia a quinolonas e integrones similares a los de patógenos humanos, sugiriendo comunicación genética entre estas bacterias de diversos ambientes. Alrededor de los recintos de acuicultura, este uso exagerado de antimicrobianos contamina con ellos también a peces silvestres para consumo humano y potencialmente selecciona BRA en su carne y en los productos de acuicultura. El consumo de estos productos selecciona bacterias resistentes en el microbioma humano y facilita también el intercambio genético entre bacterias del ambiente acuático y la microbiota comensal y patógena humana. El pasaje de antimicrobianos al ambiente marino disminuye la diversidad en él, y potencialmente podría facilitar la aparición de florecimientos de algas nocivas, la infección de peces por patógenos piscícolas resistentes los antimicrobianos y la aparición de patógenos zoonóticos resistentes, incluyendo a Vibrio parahaemolyticus. Estos hallazgos sugieren que la prevención de infecciones en peces y la disminución del uso de antimicrobianos en su crianza, será en Chile un factor determinante en la prevención de infecciones humanas y animales con resistencia múltiple a los antimicrobianos, de acuerdo con el paradigma moderno e integral de Una Salud.


The emergence and dissemination of antimicrobial-resistant bacteria (ARB) is currently seen as one of the major threats to human and animal public health. Veterinary use of antimicrobials in both developing and developed countries is many-fold greater than their use in human medicine and is an important determinant in selection of ARB. In light of the recently outlined National Plan Against Antimicrobial Resistance in Chile, our findings on antimicrobial use in salmon aquaculture and their impact on the environment and human health are highly relevant. Ninety-five percent of tetracyclines, phenicols and quinolones imported into Chile between 1998 and 2015 were for veterinary use, mostly in salmon aquaculture. Excessive use of antimicrobials at aquaculture sites was associated with antimicrobial residues in marine sediments 8 km distant and the presence of resistant marine bacteria harboring easily transmissible resistance genes, in mobile genetic elements, to these same antimicrobials. Moreover, quinolone and integron resistance genes in human pathogens isolated from patients in coastal regions adjacent to aquaculture sites were identical to genes isolated from regional marine bacteria, consistent with genetic communication between bacteria in these different environments. Passage of antimicrobials into the marine environment can potentially diminish environmental diversity, contaminate wild fish for human consumption, and facilitate the appearance of harmful algal blooms and resistant zoonotic and human pathogens. Our findings suggest that changes in aquaculture in Chile that prevent fish infections and decrease antimicrobial usage will prove a determining factor in preventing human and animal infections with multiply-resistant ARB in accord with the modern paradigm of One Health.

ABSTRACT

Acne vulgaris is a very common condition affecting up of about 80% to 90% of adolescents. The patients with acne have been shown to be adversely impacted by the effect of acne on their quality of life. Four factors are believed to play a key role in the development of acne lesions: excess sebum production, disturbed keratinization within the follicle, colonization of the pilosebaceous duct by Propionibacterium acnes, and the release of inflammatory mediators into the skin. Consequently, the target for acne therapy is these well-known pathogenic factors responsible for this disease state. Topical retinoids correct abnormal keratinization, but it should be applied cautiously because of irritation. Benzoyl peroxide is an effective bactericidal agent against P. acnes. Main topical antibiotics are erythromycin and clindamycin. Fixed combination topical products with retinoids, benzoyl peroxide and antibiotics have been introduced. Use of systemic antibiotics, including tetracyclines and macrolides rapidly improves inflammatory acne lesions. Oral isotretinoin is effective against all of the main pathogenic features of acne but is contraindicated in pregnant women and has been associated with cheilitis and dry skin. Hormonal therapy has been found to improve acne in some selective patients and should be considered for appropriate candidates. This review will present the general aspects of the pharmacological treatments for acne.

ABSTRACT Recent studies have shown a role of intestinal microbiota in obesity. The consumption of antibiotics in the last 70 years has led to changes in intestinal microbiota, which has led to weight gain and body fat accumulation. To evaluate the possibility of weight gain induced by antibiotics and the possible protective effect of probiotics, we divided 45 animals (Rattus norvegicus) into groups and administered the following treatments over two weeks: tetracycline, tetracycline + Lactobacillus gasseri, and NaCl. The animals were weighed over the course of 8 weeks, and at the end of the treatment period, they were measured and subjected to bioelectrical impedance analysis. The results show that the group receiving tetracycline alone had a higher body mass index (p=0.030), a greater Lee index (p=0.008), and a lower body water percentage than the control group, indicating a greater accumulation of body fat. The group receiving the probiotics with tetracycline presented similar results to the control group, indicating a possible protective effect of body fat accumulation in the group receiving tetracycline alone. The results show that tetracycline increased the concentration of body fat, and the use of probiotics was associated with an ability to protect the animals from the pro-obesity effect.

ABSTRACT

This study aimed at identifying the commonly screened antibiotics during the dairy processing routine in raw-milk receiving points in plants inspected by the official services in the state of Rio Grande do Sul (RS), from January 2014 to February 2015. Among the 36 participating industries, the most commonly screened antibiotics were beta lactams (100%) and tetracyclines (69%). The antibiotics screened at the milk receiving point were chosen because of the practicality and speed in performing the screening (67%), rather than specific knowledge on which antibiotics the milk suppliers used. Este estudo teve por objetivo identificar os antibióticos comumente investigados durante a rotina de processamento do leite cru recebido em laticínios sob Inspeção Estadual no Rio Grande do Sul (RS), entre janeiro de 2014 e fevereiro de 2015. Entre as 36 indústrias participantes, os antibióticos mais comumente investigados foram beta-lactâmicos (100%) e tetraciclinas (69%). A seleção por quais antibióticos investigar no recebimento do leite foi influenciada pela praticidade e rapidez na execução da análise (67%), em detrimento do conhecimento específico sobre quais antibióticos eram utilizados pelos produtores de leite (22%).(AU)

ABSTRACT

Mycoplasma pneumoniae pneumonia (MPP) is one of the most common forms of community-acquired pneumonia in children and adolescents. Outbreaks of MPP occur in 3- to 7-year cycles worldwide; recent epidemics in Korea occurred in 2006–2007, 2011, and 2015–2016. Although MPP is known to be a mild, self-limiting disease with a good response to macrolides, it can also progress into a severe and fulminant disease. Notably, since 2000, the prevalence of macrolide-resistant MPP has rapidly increased, especially in Asian countries, recently reaching up to 80%–90%. Macrolide-resistant Mycoplasma pneumoniae (MRMP) harbors a point mutation in domain V of 23S rRNA with substitutions mainly detected at positions 2063 and 2064 of the sequence. The excessive use of macrolides may contribute to these mutations. MRMP can lead to clinically refractory pneumonia, showing no clinical or radiological response to macrolides, and can progress to severe and complicated pneumonia. Refractory MPP is characterized by an excessive immune response against the pathogen as well as direct injury caused by an increasing bacterial load. A change of antibiotics is recommended to reduce the bacterial load. Tetracyclines or quinolones can be alternatives for treating MRMP. Otherwise, corticosteroid or intravenous immunoglobulin can be added to the treatment regimen as immunomodulators to down-regulate an excessive host immune reaction and alleviate immune-mediated pulmonary injury. However, the exact starting time point, dose, or duration of immunomodulators has not been established. This review focuses on the mechanism of resistance acquisition and treatment options for MRMP pneumonia.

ABSTRACT

ABSTRACT PURPOSE: To investigate the regulatory roles of neutrophil elastase (NE) and matrix metalloproteinase-9 (MMP-9) in lipopolysaccharide (LPS)-induced acute lung injury (ALI) in mice. METHODS: To construct LPS-induced ALI mouse models, wild-type C57BL/6 mice were administered 5.0 mg/kg of LPS through endotracheal, and/or 1.0 mg/kg of ONO-5046, and/or 20.0 mg/kg of chemically modified tetracycline-3 (CMT-3) by gavage. The levels of MMP-9, tissue inhibitor of metalloprotease-1, interleukin (IL)-6 were detected by real time RT-PCR at 6 h, 24 h and 48 h, and tumor necrosis factor (TNF), lung wet-dry weight ratio, white blood cell (WBC) count and polymorphonuclear (PMN) count in bronchoalveolar lavage fluid (BALF) were tested at 48 h after administration. The 5-day survival analysis of the ALI mice was also performed. RESULTS: Both ONO-5046 and CMT-3, regardless of being used individually or combined, significantly reduced the levels of MMP-9, IL-6, and TNF in lung tissue as well as in BALF, and the WBC and PMN count in BALF. Combined treatment with ONO-5046 and CMT-3 remarkably improved the survival rate of ALI mice. CONCLUSION: Neutrophil elastase synergizes with matrix metalloproteinase-9 to promote and regulate the release of inflammatory mediators and the infiltration of inflammatory cells, consequently affecting the survival of lipopolysaccharide-induced acute lung injury mice.

ABSTRACT

<p><b>BACKGROUND</b>Tetracycline (TET) has been found to have both antibiotic and anti-inflammatory properties. The anti-inflammatory effect of topical TET on atopic dermatitis (AD) has not been reported. The purpose of this study was to explore the potential role of topical TET and its anti-inflammatory effects in a mouse model of AD.</p><p><b>METHODS</b>The 2% TET was applied topically to ears of MC903-induced AD-like BALB/c mice once a day. AD-like symptoms and severity were evaluated by assessing skin scoring of dermatitis, ear thickness, and frequency of scratching. Serum IgE and thymic stromal lymphopoietin (TSLP) levels were measured by enzyme-linked immunosorbent assay. Western blot was used for analyzing the expressions of TSLP, protease-activated receptor 2 (PAR2), and nuclear factor-kappa B (NF-κB) in skin lesions. Real-time polymerase chain reaction was performed to assess the mRNA levels of TSLP and inflammatory cytokines including interleukin (IL)-4, IL-13, tumor necrosis factor (TNF)-α, and IL-1β in skin lesions.</p><p><b>RESULTS</b>Scoring of dermatitis (9.00 ± 0.63 vs. 6.67 ± 1.03, P = 0.001), ear thickness (0.44 ± 0.02 mm vs. 0.40 ± 0.03 mm, P = 0.018), and serum IgE level (421.06 ± 212.13 pg/ml vs. 244.15 ± 121.39 pg/ml, P = 0.047) were all improved in the 2% TET treatment group compared with AD group. Topical TET significantly reduced the serum level of TSLP (119.04 ± 38.92 pg/ml vs. 65.95 ± 54.61 pg/ml, P = 0.011) and both mRNA and protein expressions of TSLP in skin lesions compared with AD group (P = 0.003 and 0.011, respectively), and NF-κB and PAR2 expression in skin lesions were also suppressed (P = 0.016 and 0.040, respectively). Furthermore, expressions of inflammatory cytokines IL-4, IL-13, and TNF-α in skin lesions were down-regulated in 2% TET group compared with AD group (P = 0.035, 0.008, and 0.044, respectively).</p><p><b>CONCLUSIONS</b>Topical TET exerted anti-inflammatory effects through suppression of TSLP and inflammatory cytokines in AD mouse model, suggesting TET as a potential agent for the topical treatment of AD in the future.</p>

ABSTRACT

BACKGROUND: While 7.6% of cultured genital Mycoplasmataceae was identified as Ureaplasma urealyticum, most of them were Ureaplasma parvum (80.3%). This is the first study differentiating between these two species. We investigated the prevalence and antimicrobial resistance of genital Mycoplasmataceae in Korean women. METHODS: A total of 150 specimens submitted to the laboratory for culture of M. hominis and Ureaplasma spp. were included. Detection and antimicrobial susceptibility tests were performed with the Mycoplasma IST2 kit (bioMérieux, France). The identification of Ureaplasma spp. was performed by PCR, and mutations in drug resistance genes were investigated by PCR and sequencing. RESULTS: In total, 66 specimens (44.0%) were positive for genital Mycoplasmatacea: U. parvum, 53 (80.3%); U. urealyticum, 5 (7.6%); M. hominis, 2 (3.0%); mixed infection, 6 (9.1%). Susceptibilities of Ureaplasma spp. to erythromycin, azithromycin, clarithromycin, and doxycycline were 86.0%, 80.7%, 98.2%, and 94.7%, respectively. The susceptibility of Ureaplasma spp. to ofloxacin and ciprofloxacin was 47.4% and 17.5%, respectively. The S83L mutation was found in the ParC subunit of the ofloxacin-resistant (5/7, 71.4%) and the ciprofloxacin-resistant isolates (7/14, 50.0%). One M. hominis isolate showed resistance to erythromycin, azithromycin, and clarithromycin but susceptibility to josamycin, pristinamycin, fluoroquinolones, and tetracyclines. CONCLUSION: The prevalence of genital Mycoplasmataceae in Korean women was 44.0%; most of them were identified as U. parvum. As more than 10% of Ureaplasma spp. showed non-susceptibility to erythromycin and azithromycin (15.5%, 20.7%), a susceptibility test is needed prior to use of these antibiotics. Further study is needed about the clinical features of infections caused by U. urealyticum vs. U. parvum and their associated resistance mechanisms.

ABSTRACT

PURPOSE: The detection of high-level tetracycline-resistant strains of Neisseria gonorrhoeae (TRNG) can make important epidemiological contributions that are relevant to controlling infections from this pathogen. In this study, we aimed to determine the incidence of TRNG isolates over time and also to investigate the characteristics and genetic epidemiology of these TRNG isolates in Korea. MATERIALS AND METHODS: The antimicrobial susceptibilities of 601 isolates of N. gonorrhoeae from 2004 to 2011 were tested by standard Clinical and Laboratory Standards Institute methods. To determine the molecular epidemiological relatedness, N. gonorrhoeae multi-antigen sequence typing was performed. RESULTS: The incidence of TRNG increased from 2% in 2004 to 21% in 2011. The minimum inhibitory concentration distributions of ceftriaxone and susceptibility of ciprofloxacin in TRNG were different from non-TRNG and varied according to the year of isolation. Most of the TRNG isolates collected from 2004 to 2007 exhibited genetic relatedness, with sequence type (ST) 1798 being the most common. From 2008 to 2011, the STs of the isolates became more variable and introduction of genetically unrelated TRNG were noted. CONCLUSION: The increased incidence of TRNG strains until 2007 appears to be due, at least in part, to clonal spread. However, we propose that the emergence of various STs since 2008 could be associated with foreign import.

Las enfermedades periodontales son consecuencia de la acumulación de la placa dental. Las bacterias presentes en ella inician un proceso inflamatorio en los tejidos periodontales por medio de la liberación de toxinas bacterianas. El tratamiento indicado implica terapias mecánicas no quirúrgicas y quirúrgicas y, en algunos casos, terapia farmacológica. En pacientes que no responden a la terapia mecánica, los estudios sugieren el uso de terapias complementarias con antibióticos locales o sistémicos. En esos casos, es necesario el desarreglo previo de la placa dental adherida a la superficie radicular. ABSTRACT

<p><b>OBJECTIVE</b>To understand the antibiotic and sex hormone residue level in the Eriocheir sinensis sold in markets in Shanghai.</p><p><b>METHODS</b>Fifty six mitten crabs were collected through convenient sampling in Xuhui and Changning districts in Shanghai in 2014. The residues of 13 antibiotics (4 quinolones, 3 sulfonamides, 2 tetracyclines, 2 amphenicols, 1 macrolides, and trimethoprim) and 7 sex hormones (4 endogenous sex hormones and 3 synthetic sex hormones) were detected by the isotope dilution method based on ultra-performance liquid chromatography coupled to quadrupole/time-of-flight mass spectrometry in visceral organs and muscles of crabs.</p><p><b>RESULTS</b>A total of 7 antibiotics were detected in visceral organs or muscles of crabs, including 3 quinolones (ciprofloxacin, norfloxacin, and enrofloxacin), 3 sulfonamides (sulfamethazine, sulfadiazine, and sulfamethoxazole) and trimethoprim. In visceral organs, the overall antibiotic detection rate was 32.1%, the maximum sum of mass concentration was 23.8 µg/kg, and the 75th percentile was 1.45 µg/kg. In muscles, the overall antibiotic detection rate was 39.3%, the maximum sum of mass concentration was 40.5 µg/kg, and the 75th percentile was 1.75 µg/kg. No sex hormones were detected in both visceral organs and muscles.</p><p><b>CONCLUSION</b>The antibiotic and sex hormone residue level in the Eriocheir sinensis sold in markets in Shanghai did not exceed the national standard, indicating that no risk was posed to human health.</p>

ABSTRACT

The frequent occurrence of pharmaceuticals in the aquatic environment requires an assessment of their environmental impact and their negative effects in humans. Among the drugs with high harmful potential to the environment are the antibiotics that reach the environment not only, as may be expected, through the effluents from chemical and pharmaceutical industries, but mainly through the sewage and livestock; because around 25 to 75% of the ingested drugs are excreted in unchanged form after the passage through the Gastro-Intestinal Tract. Tetracycline has high world consumption, representing a human consumption of about 23 kg/day in Brazil in 2007. At the moment, researches are being made to develop new tetracycline that incorporate heavy metals (Hg, Cd, Re, Pt, Pd) to their structures in order to increase their bactericidal effect. The conventional wastewater treatment plants are not able to degrade complex organic molecules to reduce their toxicity and improve their biodegradability. For this reason new technologies, i.e., the advanced oxidation processes, are being developed to handle this demand. The objectives of this study are to review the literature on the processes of obtaining tetracycline, presenting its waste treatment methods and evaluation of their environmental impact.


A ocorrência frequente de fármacos no meio aquático exige a avaliação do seu impacto ambiental e seus efeitos negativos em seres humanos. Dentre os fármacos com maior potencial de impacto ambiental estão os antibióticos, que chegam ao meio ambiente através dos efluentes de indústrias químico-farmacêuticas e, principalmente, através de esgotos domésticos e criação de animais, visto que 25% a 75% dos fármacos são excretados em forma inalterada após passagem pelo Trato Gastrointestinal. Parcela significativa do consumo mundial dos antibióticos corresponde à classe das tetraciclinas, representando consumo humano de 23 kg/dia no Brasil, em 2007. Atualmente, há pesquisas de novas tetraciclinas que incorporam metais pesados (Hg, Cd, Re, Pt, Pd) às suas estruturas com o intuito de aumentar suas atividades bactericidas. As estações de tratamento de águas residuais convencionais não são capazes de degradar moléculas orgânicas complexas, diminuir a sua toxicidade e melhorar a sua biodegradabilidade. Por esta razão, as novas tecnologias, como, por exemplo, os processos oxidativos avançados, estão sendo desenvolvidos para lidar com esta demanda. Os objetivos deste trabalho são fazer uma revisão da literatura sobre os processos de obtenção de tetraciclinas, apresentar métodos de tratamento de seus resíduos e avaliar o seu impacto ambiental.

Antecedentes: En Colombia, la vigilancia del cumplimiento de los Límites Máximos de Residuos LMRs de antimicrobianos veterinarios en carne bovina es exigua, debido a que la capacidad analítica es insuficiente para monitorear un gran número de muestras y un amplio rango de antibióticos. El bioensayo es un método microbiológico desarrollado por la agencia de salud pública (FSIS) del Departamento de Agricultura de Estados Unidos, para la detección, identificación y semicuantificación de residuos antimicrobianos. Objetivos: Se evaluó preliminarmente el bioensayo para determinar su uso como prueba de detección cualitativa para el monitoreo y vigilancia de residuos de cuatro antimicrobianos en músculo bovino, como contribución al sistema de gestión de inocuidad en el marco del Acuerdo sobre la aplicación de Medidas Sanitarias y Fitosanitarias, bajo lineamientos de la Comisión Europea, Decisión 2002/657/CE. Métodos: El protocolo de experimentación usó tejidos fortificados con diferentes concentraciones de penicilina G potásica, oxitetraciclina, eritromicina y estreptomicina, como representantes de cuatro familias de antibióticos de uso veterinario en Colombia. Para el establecimiento de los criterios de funcionamiento del bioensayo se prepararon seis repeticiones de cada uno de los niveles o concentraciones probadas por antimicrobiano, incluyendo un nivel sin fortificación y cinco niveles de concentraciones antibióticas menores, iguales y superiores al Límite Máximo de Residuos LMRs de cada uno de los antimicrobianos analizados. Resultados: Se establecieron preliminarmente los criterios de funcionamiento del método (especificidad, límites de detección y estabilidad). La técnica mostró excelente especificidad (ningún falso-positivo); los límites de detección se determinaron para los cuatro antibióticos con relación a sus respectivos LMRs: Betalactámicos (penicilina G =LMR), tetraciclinas (oxitetraciclina =LMR), macrólidos (eritromicina =LMR) y aminoglucósidos (estreptomicina >4 LMR); sin embargo, la sensibilidad del bioensayo para estreptomicina no fue satisfactoria. La estabilidad de los analitos en la matriz fortificada al límite de detección, resultó adecuada durante el período de tiempo evaluado. Conclusiones: El método del bioensayo preliminarmente evaluado es aplicable a la detección de una amplia gama de residuos antimicrobianos en músculo bovino, a concentraciones iguales a los LMRs.


Background: In Colombia, the vigilance of the Maximum Residue Limits MRLs of veterinary antimicrobials in cattle meat is limited, because the analytical capacity is insufficient to monitor a large number of samples and a wide range of antibiotics. The bioassay is a microbiological method developed by the health public agency (FSIS) of the United States Department of Agriculture (USDA) for the detection, identification and semiquantification of antimicrobial residues in cattle muscle. Objective: It was evaluated preliminary the bioassay for use as a test for the qualitative detection for monitoring and surveillance of four antimicrobials in cattle muscle, as contribution to the inocuity management system within the agreement framework about the application of Sanitary and Phytosanitary Measures; using some guidelines of the European Decision 2002/657/EC. Methods: The experimental protocol used fortified tissues with varying concentrations of penicillin G potassium, oxytetracycline, erythromycin and streptomycin, as representatives of the four important families of veterinary antibiotics used in Colombia. To establish the performance criteria of the bioassay were prepared six replicates of each of the levels or concentrations tested for antimicrobial, including a level without fortification and five levels of lower antibiotic concentrations, equal and above MRL Maximum Residue Limit for each one of the tested antimicrobials. Results: The performance characteristics of method were preliminarily determined (specificity, detection limits and stability). The test showed excellent specificity (no false positive); detection limits were determined for the four antibiotics in relation to their respective Maximum Residue Limit MRLs: Beta-lactams (penicillin G =MRL), tetracyclines (oxytetracycline =MRL), macrolides (erythromycin =MRL) and aminoglycosides (streptomycin >4 MRL); however, the sensitivity of bioassay for the streptomycin was unsatisfactory. Stability of the analytes in the matrix was adequate during the period evaluated. Conclusions: The preliminary bioassay method evaluated is applicable to the detection of a broad range of antimicrobial residues in bovine muscle at concentrations at the MRLs.
